The short version

Every forum post about this stack quotes the same ratio: 100mg of l-theanine to 50mg of caffeine, one part stimulant to two parts amino acid. It is oddly specific for folk wisdom, and it is specific because it comes from an actual experiment — a 2008 crossover trial in 27 people that compared 50mg of caffeine with and without 100mg of l-theanine.
That trial is real and its result is real. It is also one study, funded by Unilever's research arm, and almost everything interesting about this combination has happened since. So here is the whole file, including the parts the ratio does not survive intact.
What the two 2008 trials found
The 50/100 study measured word recognition, rapid visual information processing, critical flicker fusion, attention switching and mood at 60 and 90 minutes after each dose. Caffeine alone improved subjective alertness at 60 minutes and attention-switching accuracy at 90. The combination improved both speed and accuracy on attention switching at 60 minutes, and reduced susceptibility to distracting information in the memory task at both time points.
A second trial the same year, at Northumbria University, used much larger doses — 250mg of theanine and 150mg of caffeine — in isolation and together. The combination improved rapid visual information processing accuracy, mental fatigue ratings, simple reaction time, numeric working memory reaction time and sentence verification accuracy.
The detail that never makes it into the stack advice: theanine on its own, in that trial, increased headache ratings and reduced the number of correct serial-seven subtractions. It is not an inert calming agent. It did something, and one of the things it did was mildly unhelpful.
The moderator analysis nobody quotes
In 2014 a meta-analysis pooled eleven randomised placebo-controlled trials of these tea constituents. It found moderate effect sizes in favour of the combination in the first two hours post-dose, on alertness ratings and on attention-switching accuracy, with smaller effects on some other attention measures. That is a fair summary of the upside and it is the best evidence the stack has.
Then the authors did a moderator analysis on the doses. Across the studies, the change in effect size tracked caffeine dose more than theanine dose, particularly in the first hour. That is not proof that theanine contributes nothing. It is a strong hint about which half of your capsule is doing the visible work.
A much larger 2025 systematic review — 50 randomised trials, 15 poolable — put numbers on the combination against placebo in healthy people: attention-switching accuracy at two hours SMD 0.33 (95% CI 0.13 to 0.54), digit vigilance accuracy 0.20 (0.02 to 0.38), choice reaction time in the first hour -0.48 (-1.01 to 0.05), overall mood 0.26 (-0.10 to 0.63). Two of those four confidence intervals cross zero. The reviewers say so themselves: the intervals frequently highlight the uncertainty around the direction and size of the difference.
The trial that ran the ratio at tea strength
This is the awkward one. A 2015 crossover study gave 12 habitual and 12 non-habitual caffeine consumers 75mg of caffeine, 50mg of theanine, both together, and placebo, measuring cerebral blood flow by near-infrared spectroscopy alongside cognition and mood.
Caffeine alone did what caffeine does: improved attention task performance and lifted overall mood, with the expected vascular changes. Adding theanine eliminated the vasoconstriction — and also eliminated caffeine's effects on cognition and mood. The authors' conclusion is blunt: at levels and ratios equivalent to one or two cups of tea, combining theanine with caffeine removed the behavioural effects of caffeine, and did not lead to a positive impact on behaviour.
So the same pairing improved attention switching at 50/100 in one trial and blunted caffeine's benefits at 75/50 in another. Dose and ratio are not decoration here. They appear to determine whether you get a lift, a smoothed lift, or a cancelled one.
What this stack does not fix
Three things, and they are the three reasons people usually go looking for it.
It does not fix caffeine tolerance. Every trial above dosed people acutely, most after a washout, and none of them tested whether a daily user gets the same effect on day ninety.
It does not fix sleep debt. Chronic restriction to six hours a night produces attention deficits comparable to two consecutive nights of no sleep at all, and no acute stack has been shown to return that. Caffeine is a mask on that curve, not a repair.
And it does not accumulate. This is an effect measured at 60 and 90 minutes and pooled over the first two hours. It is the opposite kind of intervention from something like bacopa monnieri, which shows nothing until about twelve weeks. Do not judge the two on the same timescale, and do not expect the fast one to become the slow one if you keep taking it.
What we'd actually tell you
If you already drink coffee and you like it, the honest starting position is that you are getting most of the measured effect already. Adding theanine may take the edge off; at tea-strength ratios it may also take the lift off. That is a real trade you should make deliberately rather than by copying a ratio off a forum.
If you want to test it, use the dose pair an actual trial used rather than an invented one — 50mg caffeine with 100mg theanine, or 150 with 250 — take it on a day you have a genuinely demanding task, and pay attention to task-switching and distraction rather than to how awake you feel. Those are the outcomes that moved.

Good questions
What is the best ratio of caffeine to l-theanine?
Use a pairing an actual trial used: 50mg caffeine with 100mg theanine, or 150mg with 250mg. The popular 1:2 ratio comes from the first of those. A 2015 study at 75mg caffeine and 50mg theanine found the combination cancelled caffeine's cognitive and mood effects, so the ratio is not cosmetic.
Does l-theanine actually do anything on its own?
Very little that helps. In a 2008 trial 250mg of theanine alone increased headache ratings and reduced correct serial-seven subtractions, and a 2025 meta-analysis found only a small effect on choice reaction time in the first hour. Almost all the interesting findings involve theanine combined with caffeine, not theanine by itself.
Will l-theanine stop caffeine making me jittery?
Possibly, and it may take the benefit with it. The 2015 trial that measured blood flow found theanine removed caffeine's vasoconstriction and also removed its effects on attention and mood. If jitteriness is your problem, drinking less caffeine is the more reliable fix and it costs nothing.
How long does the caffeine and l-theanine effect last?
Hours. Every trial measured at 60 and 90 minutes, and the meta-analysis pooled the first two hours post-dose. Nothing accumulates. This is the opposite of an ingredient like bacopa, which shows nothing before about twelve weeks, and judging either on the other's timescale will mislead you.
Does the stack work if I already drink coffee every day?
Unknown, and that is the honest answer. Every trial dosed people acutely, most after a washout period, and none tested a daily user at day ninety. Since the moderator analysis suggests caffeine dose drives most of the measured effect, a tolerant regular drinker has the least to gain.
Is this worth buying as a supplement?
Only if you would take the caffeine anyway and want to test the pairing deliberately. The effect is real, small, and measured in the first two hours. If you are reaching for it every afternoon, the problem is sleep or workload rather than a missing amino acid, and no capsule fixes either.
