The short version

The pitch is that theanine takes the edge off coffee — same alertness, none of the jitter. It is one of the few supplement claims with enough trials behind it to check properly, and checking it produces a more interesting answer than either the fans or the sceptics give you.
Start with the study that made the reputation
Haskell and colleagues, in Biological Psychology in 2008, ran a randomised, placebo-controlled, double-blind, balanced crossover. Every participant took each of four conditions: 250mg L-theanine alone, 150mg caffeine alone, both together, and placebo. Salivary caffeine was monitored to confirm compliance.
Theanine on its own did badly. It increased headache ratings and reduced the number of correct serial seven subtractions — that is, participants got worse at a mental arithmetic task. Caffeine on its own did what caffeine does: faster digit vigilance reaction time, better accuracy on a rapid visual information processing task, less self-reported mental fatigue.
The combination did more than caffeine alone. On top of the accuracy and fatigue improvements, it produced faster simple reaction time, faster numeric working memory reaction time and better sentence verification accuracy. Headache and tiredness ratings fell, alertness ratings rose, and there was a significant interaction between the two compounds on delayed word recognition speed.
Read the outcome list. Reaction time, accuracy, working memory, vigilance. Almost all of it is attention. The one compound that made a cognitive measure worse was theanine on its own.
What happens when you pool everything
Two teams have now done that, from opposite directions, and they agree more than you would expect.
Payne and colleagues, in Nutrition Reviews in 2025, reviewed 50 randomised trials of tea, theanine, or theanine plus caffeine in healthy participants, 15 of which could be meta-analysed. In the second hour after intake, theanine plus caffeine beat placebo on attention switching task accuracy (SMD 0.33, 95% CI 0.13 to 0.54) and digit vigilance accuracy (SMD 0.20, 95% CI 0.02 to 0.38). Two other outcomes pointed the right way but their confidence intervals crossed zero: choice reaction time in the first hour (SMD −0.48, 95% CI −1.01 to 0.05) and overall mood in the second hour (SMD 0.26, 95% CI −0.10 to 0.63). The authors note directly that the confidence intervals frequently highlight uncertainty about the direction and size of these differences. The lead author's studentship is funded by a tea company and two co-authors work for tea companies, which is disclosed.
Gerolymos and colleagues, in Molecular Psychiatry in 2026, pooled 31 randomised trials, 1,168 participants, theanine against placebo in both healthy and clinical populations. And then the honest part: the reduction in acute stress was modest, SMD 0.31, and largely influenced by studies at high risk of bias. There was no significant effect on fatigue. Anxiety effects were inconsistent and non-significant except in one trial in a clinical population. No serious adverse events were reported in any of the 31 trials.
So what is actually established
Two independent meta-analyses, different teams, different inclusion criteria, different funding pressures, both landing on attention as the robust outcome and both hedging on mood and stress. That convergence is worth more than either paper alone.
What is established: 200mg of theanine, half an hour to an hour before you need it, measurably sharpens choice reaction time, and paired with caffeine it improves accuracy on attention tasks in the first two hours. What is not established: that it makes you calmer in any durable sense. The stress signal exists, it is small, and it leans on the weakest studies in the set.
What we'd actually tell you
We do not sell theanine, so this costs us nothing to say. If you already drink coffee and you want the pairing, 200mg of theanine with your usual cup is the tested arrangement, and you should expect a sharper hour rather than a calmer day. It is inexpensive and the safety record across 31 trials is clean.
What we would not do is buy it as an anxiety product. Every effect described above is measured in hours. If your problem is a stress load that persists across weeks, this is the wrong tool, and the honest comparison is unflattering — slow breathing has been tested over months, costs nothing, and is pooled across more trials. Rhodiola sits in similar territory: another compound whose controlled evidence is about attention under load rather than about feeling different.
Good questions
How much L-theanine should I take with coffee?
200mg, taken 30 to 60 minutes before you need the effect, is the dose with the cleanest meta-analytic support. The classic combination trial used 250mg of theanine with 150mg of caffeine, roughly a strong cup. Higher doses have not been shown to do more, and the measured benefits are in attention rather than calm.
Does L-theanine stop caffeine jitters?
The evidence for that specific claim is weaker than its popularity suggests. Pooled analyses find the reliable effect is on attention measures, while mood and stress outcomes have confidence intervals crossing zero or lean on high-risk-of-bias trials. In the main crossover study, headache and tiredness ratings did fall with the combination, so there is something there — just less than advertised.
Can I take L-theanine on its own without caffeine?
You can, and one trial found it made a mental arithmetic task worse and increased headache ratings when taken alone. The 2026 pooled analysis still found a solo 200mg dose improved choice reaction time, so it is not inert. But the combination is where the more consistent cognitive data sits, and theanine alone is not a sedative.
Will L-theanine help my anxiety?
Probably not in any way you would notice. Across 31 randomised trials, anxiety effects were inconsistent and non-significant apart from one study in a clinical population. Every effect in this literature is measured in hours, not weeks. If you are dealing with persistent anxiety, that is a conversation with a doctor rather than a supplement purchase.
Is L-theanine safe to take every day?
The short-term record is good — no serious adverse events across 31 randomised trials, and the mild effects reported were things like headache. What does not exist is long-term data, because almost every trial dosed acutely or over a few weeks. Nobody has followed daily users for a year, and we are not going to describe that silence as reassurance.
