The short version
- Much of the L-arginine you swallow is destroyed by intestinal arginase before it reaches circulation, which is why oral L-citrulline raised plasma arginine more effectively than arginine itself in a 20-person crossover trial.
- Pooling 12 studies and 198 participants, at least 3g of citrulline taken at least 30 minutes before testing produced a small but significant strength and power benefit, a standardised mean difference of 0.20.
- Citrulline malate at 6 to 8g taken 40 to 60 minutes before training added about 3 reps, or 6.4%, versus placebo across 8 studies in 137 mostly strength-trained people.

Turn a pre-workout tub around and L-arginine is usually sitting somewhere in the middle of the panel, printed with the confidence of an ingredient that works. The logic looks airtight: arginine is the direct substrate for nitric oxide, so swallow arginine, make more nitric oxide, dilate more vessels, move more blood. The biochemistry is not the problem. The digestive tract standing between the capsule and your bloodstream is.
Presystemic elimination, plainly
Schwedhelm and colleagues set out to test this properly in British Journal of Clinical Pharmacology in 2008. Their framing of the problem is blunt: oral arginine treatment is hampered by extensive presystemic elimination due to intestinal arginase activity, while citrulline is readily absorbed and converted, at least in part, to arginine downstream. In plain terms, a large share of the arginine you swallow is broken down before it ever counts.
They ran a double-blind, randomised, placebo-controlled crossover in 20 healthy volunteers across six dosing regimens, each for a week. The headline result: L-citrulline raised plasma L-arginine — both peak concentration and total exposure — more effectively than L-arginine itself, dose-dependently, at p < 0.01. Citrulline is a better way to raise your arginine than arginine is. That one sentence is the whole case for the ingredient.
The downstream markers followed. At the top dose of 3g twice daily, the arginine-to-ADMA ratio rose from 186 to 278 (p < 0.01), urinary nitrate went from 92 to 125 µmol/mmol creatinine (p = 0.01), and urinary cGMP from 38 to 50 nmol/mmol creatinine (p = 0.04).
And the honest bit: no treatment improved flow-mediated dilation over baseline. The signalling markers moved. The vascular function endpoint the authors were hoping for did not — though a pooled analysis of all the flow-mediated dilation data did show a correlation between the rise in the arginine/ADMA ratio and improvement in dilation. If you take one caveat from this article, take that one.
Does it show up in the set?
Trexler and colleagues meta-analysed this in Sports Medicine in 2019, and their inclusion criteria are why the number is trustworthy: at least 3g of citrulline, given at least 30 minutes before testing, with studies excluded if citrulline was combined with creatine, caffeine, nitrate or other ergogenic ingredients. Twelve studies, 13 samples, 198 participants. The result was a significant benefit over placebo (p = 0.036) with a pooled standardised mean difference of 0.20, 95% CI 0.01 to 0.39. Heterogeneity was essentially zero and there was no funnel plot asymmetry — a tidy analysis reporting a small effect.
Vårvik and colleagues asked the narrower question in 2021: does citrulline malate buy you more reps before failure? Eight studies, 137 participants, mostly strength-trained men. At 6–8g taken 40–60 minutes before training, it increased repetitions by 3 ± 5 reps, or 6.4 ± 7.9%, versus placebo (p = .022, SMD 0.196). Then the subanalyses: lower body 8.1% (p = .051), upper body 5.7% (p = .131). Neither reached significance on its own.
Read those two together and you have the truth of this ingredient. The pooled effect is real, it is small, and it does not survive being cut in half. That is not a reason to skip it. It is a reason to know what you bought.
The stack question
If citrulline beats arginine, why would anyone put both in a product? Because a small body of work suggests the combination beats either alone. Suzuki and colleagues reported in 2017 that 1g of citrulline plus 1g of arginine raised plasma arginine more efficiently in healthy males than 2g of citrulline or 2g of arginine on its own.
A 2019 trial in European Journal of Applied Physiology took that to a performance test. Twenty-four male collegiate soccer players took 1.2g each of citrulline and arginine daily for six days, then dosed an hour before a 10-minute full-power cycling test, in a double-blind, randomised, placebo-controlled crossover. Power output was significantly higher on the stack — 242 ± 24 W against 231 ± 21 W (p < 0.05). Post-exercise nitrite/nitrate, citrulline and arginine were all elevated, and the players reported less leg muscle soreness and greater ease of pedalling.
That is roughly a 5% power difference, and here is the disclosure that belongs next to it: the study was funded by Kyowa Hakko Bio, and three co-authors were company employees. It does not invalidate the result — the design is sound and the conflict is declared in the paper — but we sell a citrulline-arginine stack, so we are the last people who should quote that trial without saying who paid for it.
What we'd actually tell you
Match the dose to the literature or do not bother. The trials used 3g or more of citrulline, or 6–8g of citrulline malate, taken 40–60 minutes before training. If a pre-workout lists 1g of citrulline buried in a proprietary blend, it sits below every dose that produced these results — and the blend exists so you cannot check. If it lists arginine on its own, you now know what intestinal arginase does with most of it.
Set your expectations at the effect size rather than the marketing. A small standardised mean difference feels, from the inside, like a couple of extra reps across a session and a sense that the work was easier than usual. It will not add plates to your bench. It might add the last two reps of your fifth set, which across a training block is not nothing — and it is also not a transformation, which is why we are not going to call it one.
One practical note: citrulline is not a stimulant, and the meta-analysis above deliberately excluded studies pairing it with caffeine or creatine. That small effect is citrulline standing on its own. If you take it expecting the jolt of a pre-workout, you will conclude it does nothing.
Good questions
Is L-arginine a waste of money?
As a standalone way to raise your arginine, largely yes. Oral arginine is hampered by extensive presystemic elimination from intestinal arginase, and in a 20-person crossover trial citrulline raised plasma arginine more effectively than arginine did. The one argument for including both is a small body of work suggesting 1g of each beats 2g of either alone, and we sell that combination, so weigh it accordingly.
How much citrulline should I take before training?
Match the trials or do not bother. The strength and power meta-analysis required at least 3g taken at least 30 minutes before testing. The repetitions analysis used 6 to 8g of citrulline malate 40 to 60 minutes beforehand. Those are the doses that produced the published effects, and a smaller dose is not a smaller version of the result. If a label hides the amount in a blend, that is the point of the blend.
What does citrulline actually feel like?
Less than the marketing suggests. The pooled effect across 12 studies was a standardised mean difference of 0.20, which from the inside feels like a couple of extra reps across a session and a sense that the work was easier than usual. It will not add plates to your bench. Across a training block those extra reps are not nothing, but they are also not a transformation.
Is citrulline a stimulant?
No. It contains no caffeine and produces no jolt. The meta-analysis showing a benefit deliberately excluded studies that paired citrulline with creatine, caffeine or nitrate, so that small effect is citrulline standing on its own. If you take it expecting the hit of a stimulant pre-workout, you will conclude it does nothing, and you will be judging it on the wrong outcome.
Does citrulline improve blood flow or blood vessel function?
The markers moved; the function test did not. At 3g twice daily the arginine-to-ADMA ratio rose from 186 to 278 and urinary nitrate and cGMP both increased. But no treatment in that trial improved flow-mediated dilation over baseline, which was the vascular endpoint the authors were after. Anyone quoting citrulline as a proven circulation product is quoting the markers and skipping the endpoint.
Is the research on citrulline-arginine stacks independent?
Not entirely, and you should know that from us. The 2019 cycling trial showing higher power output on a combined citrulline and arginine dose was funded by Kyowa Hakko Bio, with three co-authors employed by the company.
