The short version
- A single 50,000 IU dose of vitamin D3 produced 32% higher peak plasma vitamin D3 at 12 hours when taken with a fat-containing meal rather than a fat-free one, in 50 healthy older adults.

There is a genre of morning routine video where seven bottles are lined up on a kitchen counter and each one is assigned a slot: this one on an empty stomach at 6am, this one with fat at 8, this one only after training, this one strictly at night. It looks like rigour. Most of it is invented.
Some timing rules are real and have human trials behind them. There are about four. Here they are, and then here is the honest accounting of what is left over.
Rule one: fat-soluble vitamins go with a meal that contains fat
This one is well demonstrated. Dawson-Hughes and colleagues gave 50 healthy older adults a single 50,000 IU dose of vitamin D3 with one of three breakfasts: fat-free, or 30% of calories as fat with a low or high ratio of monounsaturated to polyunsaturated fat. Peak plasma vitamin D3 at 12 hours was 32% greater (95% CI 11% to 52%) with the fat-containing meals than the fat-free one.
Interestingly, the type of fat made no difference. So the practical rule is simple: vitamins A, D, E and K with a meal that has some fat in it. Not a specific fat. Not a specific meal. Just not on an empty stomach with black coffee.
Rule two: caffeine has a longer tail than you think
Drake and colleagues gave 400mg of caffeine at bedtime, three hours before bed, and six hours before bed, against placebo, and measured sleep at home both by self-report and with a validated monitor. All three timings significantly disrupted sleep relative to placebo. The six-hour result is the one people find surprising: caffeine taken at 4pm was still measurably degrading sleep at 10pm.
This is the highest-value timing rule on the list, because the thing it protects — sleep — outperforms every supplement anyone sells, including ours.
Rule three: melatonin is a timing signal, and taking it at bedtime mostly wastes that
Melatonin is the clearest case of a supplement where when is the entire mechanism. Burgess, Revell and Eastman built a phase response curve in 27 healthy adults by giving 3mg of melatonin at fixed clock times and measuring the shift in each person's own melatonin rhythm.
The curve has structure. The phase-advance peak — the point where melatonin pulls your clock earlier — is about five hours before your own melatonin onset, which for most people lands in the afternoon. The delay peak is about eleven hours after onset, shortly after normal waking. And there is a dead zone of minimal shift across the first half of habitual sleep. The authors' own conclusion: using melatonin as a sleep aid at night "has minimal phase shifting effects."
So if you are taking melatonin to shift a body clock — jet lag, a schedule change, a late-running sleep phase — the timing is the intervention, and bedtime is the wrong slot. If you are taking it at 10pm for a mild sedative effect, fine, but understand you are using it as a different drug than the circadian literature is describing. Either way, a lower dose taken earlier is closer to what the research supports than a big dose at lights-out.
Rule four: for iron, frequency beats amount
Stoffel and colleagues ran two randomised trials in iron-depleted women using stable iron isotopes. In the first, 60mg of iron given on alternate days produced higher cumulative fractional absorption than the same doses on consecutive days — 21.8% versus 16.3% — and more total iron absorbed (175.3mg versus 131.0mg). In the second, splitting 120mg into two daily doses did not beat taking it as one morning dose.
The mechanism is hepcidin: a dose of iron raises it, and the raised hepcidin blocks absorption of the next dose. This is the clearest example of a real timing finding that runs directly against the intuitive "more often is better" rule. It is also a reminder that iron supplementation should follow a blood test, not a hunch.
What is left over
"Take magnesium at night." The sleep trials on magnesium did dose in the evening, so evening is a reasonable default by inheritance. But nobody has run the morning-versus-night comparison. It costs nothing to take it at night, so take it at night if you like — just know you are following a convention, not a finding.
"Empty stomach for maximum absorption." True for a handful of specific things. Mostly untested for the botanicals it gets applied to. And it has a real cost: on an empty stomach, more people get nausea, and nauseated people stop taking things. Adherence beats a marginal absorption difference every time.
"Creatine has an anabolic window." Creatine works by saturating muscle stores over days and weeks. Total daily intake and consistency are what move that. The clock is not doing much work here.
Elaborate multi-slot schedules generally. The more slots a protocol has, the more likely you are to miss one, and a missed dose is a bigger effect than any of these refinements.
What we'd actually tell you
Keep four rules: fat-soluble vitamins with a meal containing fat; caffeine before mid-afternoon; melatonin early and low if you are shifting a clock; iron by blood test, and alternate days rather than doubled up.
For everything else, the best time to take a supplement is the time you will actually remember. Attach it to something you already do without thinking — the coffee, the toothbrush, the dog. A perfect schedule you follow four days a week loses to a crude one you follow seven.
